"Theoretically, one should obtain essentially the same clinical picture from failure of an end-organ to respond to a hormone as from a decreased production or absence of said hormone. " With these words, Fuller Albright began his now classic paper describing a novel disease, pseudo hypoparathyroidism (PHP), and a novel concept, hormone resis- tance as a cause of disease. Soon, other hormone resistance disorders such as nephrogenic diabetes insipidus (NDI) were recognized, and the concept was extended to resistance to other substances such as calcium ions in familial hypocalciuric hypercalcemia (FHH). Later, diseases characterized by excess rather than deficient hormone action such as McCune-Albright syndrome (MAS) and familial male precocious puberty (FMPP) were recognized to be caused by autonomous endocrine hyperfunction. Although many i!!vestigators provided careful and detailed descriptions of the clinical features of these and other related endocrine disorders, an understanding of pathogenesis proved elusive for many years. In just the past few years, we have gone from clinical description to a molecular understanding of these interesting disorders. This remarkable progress reflects a synthe- sis of three distinct, but now overlapping, areas of biomedical research: the aforemen- tioned recognition and careful clinical description of specific diseases, the elucidation of the basic mechanisms of signal transduction, and the application of the powerful tools of molecular biology and genetics. Fundamental studies on the mechanisms of hormone action by Rodbell and colleagues at NIH culminated in the discovery of a major signal transduction pathway involving heterotrimeric G proteins.
G Protein Methods and Protocols
The G proteins are a family of structurally homologous, plasma membrane-associated guanine-nucleotide-binding proteins. These proteins play an integral role in the tra- duction of extracellular signals through second messenger systems. As such, G proteins affect a wide variety of intra cellular biochemical reactions by regulating the concent- tion of second messengers in cells. G proteins are heterotrimeric, consisting of a, p, and y polypeptide chains, with G protein specificity largely det- mined by the a-subunit, Molecular cloning of G protein s- units has revealed 23 distinct a-subunits, encoded by 17 different genes. Based on functional measures, G proteins are generally classified into three major categories: the G, family, which is stimulatory for adenylyl cyclase; the G, f- ily, which is inhibitory for adenylyl cyclase; and the G, f- ily, which stimulates phospholipases (Birnbaumer and Birnbaumer, 1995). Alternatively, on the basis of sequence homology, G proteins can be subdivided into four cate- ries: G,, G,, G,, and G12.

Whereas the US military and politicians hugely profited from cocaine under the guise of funding rebel armies in Central America to overthrow rogue governments, the Mujaheddin brought in profits from heroin under the guise of shutting down opium drug lords and driving out the Soviet army. Back on American soil, America turned a blind eye on her own children.G.O.D. lays out how the global greed and the shady economic practices of the rich and powerful affect the impoverished. In the book, Hamid and Luqman try to balance the tables by getting American educations. Their goal is to help their people and their motherland by waging a war of wits. Brute force is traded for intelligence. Rage is traded for strategy. Drugs in the inner city are traded for money which is used to fund their objectives.Saleem Little wrote G.O.D. in a way that was both informative and entertaining. He distilled vast amounts of information and global history into easy-to-understand paragraphs that articulated his points. In doing so, he puts the actions of his characters in proper contexts. He writes "Intellectual Urban Fiction" that has its base in the inner-city and its connections to global phenomena. Little illustrated the butterfly effects of billionaires controlling the puppet strings of governments that turn a blind eye to a profitable inter-continental drug trade. Communities sprinkled across the globe are targeted by both law enforcement agencies and criminal activities. The billionaires profit while the disadvantaged are negatively affected in numerous ways by a "War on Drugs."Although the ending of G.O.D. is tragic, there are many lessons that Saleem Little pulls into clear view. Little walks a thin line between fact and fiction with G.O.D.
