Cette tude a port sur cinq antig nes, chacun examin avec trois immunoglobulines (IgG totale, IgG1 et IgG4) l'aide de la m thode ELISA de titrage en damier afin de d terminer la concentration optimale de l'antig ne et les dilutions de s rum pour la pr paration des kits de diagnostic. La m thode ELISA indirecte a ensuite t utilis e pour valuer les valeurs de performance diagnostique (sensibilit , sp cificit , efficacit diagnostique, valeur pr dictive positive et valeur pr dictive n gative) sur la base de la valeur seuil de chaque antig ne avec l'IgG totale et ses sous-classes (IgG1 et IgG4). L'antig ne B (AgB) a t purifi partir du liquide hydatique et trait par lectrophor se pour isoler ses bandes (AgB 8 kDa, 16 kDa et 24 kDa), qui ont t utilis es comme antig nes, en plus d'un antig ne purifi partir de protoscoleces. Parmi les 15 kits pr par s, ceux contenant la sous-unit 8 kDa de l'AgB et l'IgG4 ont montr les meilleures performances. Les r sultats de la courbe ROC ont indiqu que la sous-unit 8 kDa avait la plus grande pr cision diagnostique lorsqu'elle tait test e avec les IgG4 et les IgG1. En outre, cette tude comprenait l'immunisation de lapins avec l'AgB, et les r sultats ont indiqu que l'AgB peut moduler la r ponse immunitaire vers un biais Th2.

Tra i 15 kit preparati, quelli contenenti la subunit 8 kDa di AgB e le IgG4 hanno mostrato le migliori prestazioni. I risultati della curva ROC hanno indicato che la subunit 8 kDa ha avuto la massima accuratezza diagnostica quando stata testata con IgG4 e IgG1. Antygen B (AgB) oczyszczono z plynu hydatidowego i poddano obr bce elektroforetycznej w celu wyizolowania jego pasm (8 kDa, 16 kDa i 24 kDa AgB), kt re wykorzystano jako antygeny, opr cz antygenu oczyszczonego z protoskolec w. Spośr d 15 przygotowanych zestaw w, te zawierające podjednostkę 8 kDa AgB i IgG4 wykazaly najlepszą wydajnośc. Wyniki z krzywej ROC wskazaly, że podjednostka 8 kDa miala najwyższą dokladnośc diagnostyczną, gdy byla testowana z IgG4 i IgG1.
