In The Name of Justice: The Erica Blackstone Chronicles is the spin-off of the three-book Broken Promises series. The main character Erica Blackstone former crime reporter forms a private investigation firm with Jasmine Jefferson (JJ), Jefferson and Blackstone Investigations (JBI). In their first year of partnership, they take on a child custody case of a close friend of JJ's (Benjamin Bailey). Benjamin hires JBI to investigate his soon-to-be ex-wife (Nina) to prove she is unfit to care for their two minor children. Nina will stop at nothing in her quest to get full custody of her children. The devious plan she devises is shocking and gives Benjamin proof that his wife has lost all touch with reality. As the case progresses, Erica and JJ's personal and private lives begins to spiral out of control when inappropriate pictures are sent to Erica's children's school and to JJ's husband (Lewis). On top of that, JJ's infant son (Leo) is kidnapped which leaves Erica to not only operate the firm alone, but also to assist in the search for the missing child. Added to the disappearance of Leo, Erica's, Aunt Arlene disaapears when intensity is high because of her upcoming wedding. As family and friends come together to deal with the many issues that plague them, will they be able to overcome their issues, or will they fall prey to the evil forces that are working overtime to tear them apart?

In letzter Zeit hat sich die Metagenomik als Alternative zu den herk mmlichen Methoden des mikrobiologischen Screenings herausgestellt, da sie eine umfassende Untersuchung der mikrobiellen Gemeinschaften in ihrer nat rlichen Umgebung erm glicht. Hier wurde diese Technik verwendet, um die Ausdehnung und Vielfalt der Resistenzmechanismen der mikrobiellen Gemeinschaften zu bewerten, die mit der Rhizosph re von Erica andevalensis, einer extremophilen und endemischen Heidepflanze des Rio Tinto Flusses in Spanien, assoziiert sind. Zu diesem Zweck wurde eine metagenomische Bibliothek erstellt, die auf dem Plasmid pBluescript SKII+ als Klonierungsvektor basiert und Inserts mit einer Gr e zwischen 2500 bp und 8000 bp (Basispaar) aufnehmen kann. Unsere Bibliothek umfasste 60.000 Klone, die wir auf vier Antibiotika selektierten: Nalidixins ure, Tetracyclin, Streptomycin und Kanamycin. Gegen diese isolierten wir einen resistenten Klon mit einem MIC = 10μg/ml. Die Sequenzierung des Inserts und seine anschlie ende bioinformatische Analyse ergab, dass es sich um eine Aminoglykosid-Phosphotransferase APH III handelt, die durch Deaktivierung durch Phosphorierung des Antibiotikums wirkt.

Recently, metagenomics has emerged as an alternative to conventional microbiological screening methods, since it allows comprehensive study of microbial communities in their natural environment. Here, we used this technique to assess the extent and diversity of resistance mechanisms in microbial communities associated with the rhizosphere of Erica andevalensis, an extremophilic heather endemic to the Rio Tinto river, Spain. To this end, we built a metagenomic library based on the pBluescript SKII+ plasmid as cloning vector, accepting inserts ranging in size from 2500bp to 8000bp (base pair). Our library included 60,000 clones, to which we subjected four antibiotics: nalidixic acid, tetracycline, streptomycin and kanamycin. Against these, we isolated a resistant clone with MIC = 10μg/ml. Sequencing of the insert and subsequent bioinformatics analysis revealed it to be an aminoglycoside phosphotransferase APH III, which acts by phosphorylation deactivation of the antibiotic.

Recentemente, la metagenomica emersa come un'alternativa ai metodi di screening microbiologico convenzionali, poich consente di studiare in modo esaustivo le comunit microbiche nel loro ambiente naturale. Questa tecnica stata utilizzata per valutare l'estensione e la diversit dei meccanismi di resistenza nelle comunit microbiche associate alla rizosfera di Erica andevalensis, un'erica estremofila endemica del fiume Rio Tinto, in Spagna. A tal fine, abbiamo costruito una libreria metagenomica utilizzando il plasmide pBluescript SKII+ come vettore di clonazione, accettando inserti di dimensioni comprese tra 2500 bp e 8000 bp (coppia di basi). La nostra libreria conteneva 60.000 cloni, ai quali abbiamo sottoposto quattro antibiotici: acido nalidixico, tetraciclina, streptomicina e kanamicina. Contro questi, abbiamo isolato un clone resistente con una MIC = 10μg/ml. Il sequenziamento dell'inserto e la successiva analisi bioinformatica hanno rivelato che si tratta di un'aminoglicoside fosfotransferasi APH III che agisce per disattivazione della fosforilazione dell'antibiotico.
