In letzter Zeit hat sich die Metagenomik als Alternative zu den herk mmlichen Methoden des mikrobiologischen Screenings herausgestellt, da sie eine umfassende Untersuchung der mikrobiellen Gemeinschaften in ihrer nat rlichen Umgebung erm glicht. Hier wurde diese Technik verwendet, um die Ausdehnung und Vielfalt der Resistenzmechanismen der mikrobiellen Gemeinschaften zu bewerten, die mit der Rhizosph re von Erica andevalensis, einer extremophilen und endemischen Heidepflanze des Rio Tinto Flusses in Spanien, assoziiert sind. Zu diesem Zweck wurde eine metagenomische Bibliothek erstellt, die auf dem Plasmid pBluescript SKII+ als Klonierungsvektor basiert und Inserts mit einer Gr e zwischen 2500 bp und 8000 bp (Basispaar) aufnehmen kann. Unsere Bibliothek umfasste 60.000 Klone, die wir auf vier Antibiotika selektierten: Nalidixins ure, Tetracyclin, Streptomycin und Kanamycin. Gegen diese isolierten wir einen resistenten Klon mit einem MIC = 10μg/ml. Die Sequenzierung des Inserts und seine anschlie ende bioinformatische Analyse ergab, dass es sich um eine Aminoglykosid-Phosphotransferase APH III handelt, die durch Deaktivierung durch Phosphorierung des Antibiotikums wirkt.
Recently, metagenomics has emerged as an alternative to conventional microbiological screening methods, since it allows comprehensive study of microbial communities in their natural environment. Here, we used this technique to assess the extent and diversity of resistance mechanisms in microbial communities associated with the rhizosphere of Erica andevalensis, an extremophilic heather endemic to the Rio Tinto river, Spain. To this end, we built a metagenomic library based on the pBluescript SKII+ plasmid as cloning vector, accepting inserts ranging in size from 2500bp to 8000bp (base pair). Our library included 60,000 clones, to which we subjected four antibiotics: nalidixic acid, tetracycline, streptomycin and kanamycin. Against these, we isolated a resistant clone with MIC = 10μg/ml. Sequencing of the insert and subsequent bioinformatics analysis revealed it to be an aminoglycoside phosphotransferase APH III, which acts by phosphorylation deactivation of the antibiotic.
