Typ-2-Diabetes mellitus ist eine fortschreitende Krankheit, von der fast 8,3 % der Weltbev lkerung betroffen sind. Trotz der gro en Auswahl an oralen Hypoglyk mika erreichen nur 36 % der Patienten eine angemessene Blutzuckerkontrolle. Aufgrund des geringen Hypoglyk mierisikos hat DPP-IV die Aufmerksamkeit der medizinischen Chemiker als neues Ziel f r orale Hypoglyk mika auf sich gezogen. In diesem Bericht wurde eine Leitverbindung (1) mit einem Antipyrin-Ger st erhalten, und ihr Bindungsmodus wurde berechnet. Mehrere Derivate mit verbr ckten stickstoffhaltigen Heterocyclen wurden durch eine Mehrkomponentenreaktion unter kontrollierten Mikrowellenerw rmungsbedingungen synthetisiert. Die antidiabetische Aktivit t gegen ber dem DPP-IV-Protein wurde bewertet und mit Sitagliptin verglichen. Verbindungen mit kleinen oder mittelgro en stickstoffhaltigen Br cken waren hinsichtlich der DPP-IV-Hemmaktivit t mit Sitagliptin vergleichbar, m glicherweise ber den Angriff auf Glu203 und Glu204. Die oralen hypoglyk mischen Aktivit ten der Verbindungen mit submikromolaren IC50-Werten wurden anhand eines diabetischen Mausmodells weiter untersucht.

Le diab te sucr de type 2 est une maladie progressive qui touche pr s de 8,3 % de la population mondiale. Malgr la disponibilit d'une grande vari t d'hypoglyc miants oraux, seuls 36 % des patients parviennent contr ler correctement leur glyc mie. Avec l'avantage d'un faible risque d'hypoglyc mie, la DPP-IV a attir l'attention des chimistes m dicinaux en tant que nouvelle cible pour les hypoglyc miants oraux. Dans ce rapport, un compos principal 1, avec un chafaudage antipyrine, a t obtenu et son mode de liaison a t calcul . Plusieurs d riv s avec des h t rocycles azot s pont s ont t synth tis s via une r action plusieurs composants dans des conditions contr l es de chauffage par micro-ondes. L'activit antidiab tique contre la prot ine DPP-IV a t valu e et compar e la sitagliptine. Les compos s avec des ponts azot s de taille petite ou moyenne taient comparables la sitagliptine en termes d'activit inhibitrice de la DPP-IV, potentiellement en ciblant les Glu203 et Glu204. Les activit s hypoglyc miques orales des compos s ayant des valeurs IC50 submicromolaires ont t valu es de mani re plus approfondie en utilisant un mod le de souris diab tique.

Apesar da disponibilidade de uma grande variedade de hipoglicemiantes orais, apenas 36% dos doentes conseguem um controlo glic mico adequado. Com a vantagem do baixo risco de hipoglicemia, a DPP-IV atraiu a aten o dos qu micos medicinais como um novo alvo para os hipoglicemiantes orais. Neste relat rio, foi obtido um composto principal 1, com estrutura antipirina, e o seu modo de liga o foi calculado. V rios derivados com heterociclos azotados em ponte foram sintetizados atrav s de uma rea o multicomponente em condi es controladas de aquecimento por micro-ondas. A atividade antidiab tica contra a prote na DPP-IV foi avaliada e comparada com a sitagliptina. Os compostos com pontes azotadas de tamanho pequeno ou m dio foram compar veis sitagliptina em termos de atividade inibidora da DPP-IV, potencialmente atrav s do alvo Glu203 e Glu204. As actividades hipoglic micas orais dos compostos com valores submicromolares de IC50 foram ainda avaliadas utilizando o modelo de ratinho diab tico.
