Neste estudo, a explora o mineira SNP foi realizada nas sequ ncias transcritas de M. rosenbergii dispon veis no GenBank, NCBI. Foram recolhidos esp cimes selvagens de diferentes massas de gua. Os prim rios foram concebidos para amplificar a codifica o (CDS) e sequenciaram os amplicons nas plataformas GS-FLX 454 e Ion torrent. Foram detectados 173 SNPs entre popula es. Esta abordagem para os SNPs de minera o em sequ ncias relatadas foi utilizada por n s pela primeira vez.

Die Art kann ihren fr heren bevorzugten Status wiedererlangen, wenn sie in Bezug auf Wachstum und Krankheitsresistenz genetisch verbessert wird. Die selektive Zucht wird durch die markergest tzte Selektion (MAS) erheblich erleichtert. Einzelnukleotid-Polymorphismen (SNPs) bieten die feinste Aufl sung, die f r eine Markerkarte m glich ist (eine einzelne Nukleotidver nderung), sind kodominant, bi-allelisch und in der Regel in Populationen mit niedriger Mutationsrate h ufig. Die DNA-Sequenzierung ber NGS-Plattformen gilt als der kosteng nstigste Ansatz. In dieser Studie wurde ein SNP-Mining in den transkribierten Sequenzen von M. rosenbergii durchgef hrt, die in GenBank, NCBI, verf gbar sind. Es wurden wilde Exemplare aus verschiedenen Gew ssern gesammelt. Es wurden Primer zur Amplifikation der kodierenden Sequenzen (CDS) entwickelt und die Amplikons auf GS-FLX 454 und Ion torrent Plattformen sequenziert. Es wurden 173 SNPs in den verschiedenen Populationen entdeckt. Dieser Ansatz zum Abbau von SNPs in gemeldeten Sequenzen wurde von uns zum ersten Mal verwendet.

Sono stati disegnati dei primer per amplificare la codifica (CDS) e sequenziati gli ampliconi su piattaforme GS-FLX 454 e Ion torrent. Sono stati rilevati 173 SNP nelle varie popolazioni. Questo approccio per l'estrazione di SNP nelle sequenze riportate stato utilizzato da noi per la prima volta.

Amongst farmed crustaceans, the giant freshwater prawn Macrobrachium rosenbergii was until recently one of the major species with high commercial value. Lately, it has lost popularity due to diseases and differential growth of males on account of social hierarchy. The species can reacquire its former preferred status if it is genetically improved for growth and disease resistance. Selective breeding is greatly facilitated by marker-assisted selection (MAS). Single nucleotide polymorphisms (SNPs) offer the finest resolution possible for a marker map (a single nucleotide change), are co-dominant, bi-allelic and generally abundant in populations with a low mutation rate. DNA sequencing through NGS platforms is regarded as the most cost-effective approach. In this study, SNP mining was carried out in the transcribed sequences of M. rosenbergii available in GenBank, NCBI. Wild specimens were collected from different water bodies. Primers were designed to amplify coding (CDS) and sequenced the amplicons on GS-FLX 454 and Ion torrent platforms. 173 SNPs were detected across populations. This approach for mining SNPs in reported sequences has been used by us for the first time.
