The aim of the present work was to develop and validate a simple RP-HPLC method for the determination of Rebamipide. Chromatographic separation of Rebamipide was achieved by using a C18 column. A Mobile phase containing a mixture of methanol: water (70:30 v/v) was pumped at the flow rate of 1 ml/min. Detection was performed at 231 nm. Validation parameters were evaluated according to the International Conference on Harmonization (ICH) Q2R1 guidelines. The calibration curve was linear in the concentration range 2- 10μg/ml for the Rebamipide with regression coefficient 0.999. The method was found to be accurate with 0.66 % RSD. RSD values were found to be 0.94 % in the case of intra-day precision studies, whereas0.76% in the case of inter-day precision the limits of detection and quantification were found to be 0.6, 1.98μg/ml, respectively. The results obtained from the stress testing reveals that Rebamipide drug substance is particularly stable under acidic, neutral and thermal degradation conditions. From the stability point of view, care should be taken in the manufacturing process and storage of this product

Si ritiene che le attivit dei flavonoidi aumentino quando sono coordinati con ioni metallici di transizione. Un tentativo stato fatto per capire il contributo della coordinazione del metallo e il tipo di interazione tra un flavonoide e lo ione metallico e la valutazione della sua attivit biologica. In questo studio un nuovo complesso metallico di Co(II) con la naringina stato sintetizzato e caratterizzato da FT-IR, spettrofotometro UV-VIS. Gruppi di sei topi sono stati trattati con il complesso di Cobalto Naringina (10 mg/kg di peso corporeo), somministrato per via endovenosa. Sono stati utilizzati diversi test per valutare l'effetto anti-nocicettivo o l'influenza sul comportamento delle sostanze testate sulla nocicezione termica, la nocicezione chimica e il comportamento spontaneo. Nello studio attuale stato valutato che il complesso ha uno ione Co(II) coordinato attraverso la posizione 4 e 5 del flavonoide. Il metallo si lega con il flavonoide nel rapporto 1:2, che stato confermato dal metodo Jobs. I nostri dati preliminari per le latenze di risposta per i test hot-plate e tail- flick suggeriscono che il CNC somministrato per via sistemica produce un significativo effetto analgesico sotto la stimolazione termica nocicettiva. Il test del comportamento spontaneo mostra una significativa diminuzione dell'attivit .

Uważa się, że aktywnośc flawonoid w wzrasta, gdy są one skoordynowane z jonami metali przejściowych. Podjęto pr bę zrozumienia udzialu koordynacji metali i rodzaju interakcji pomiędzy flawonoidem i jonami metali oraz oceny ich aktywności biologicznej. W niniejszej pracy zsyntetyzowano nowy kompleks Co(II) z naringiną i scharakteryzowano go za pomocą spektrofotometru FT-IR i UV-VIS. Grupom sześciu myszy podawano kompleks kobaltu z narynginą (10 mg/kg masy ciala), podawany IP. W celu oceny efektu antynocyceptywnego lub wplywu badanych substancji na zachowanie zastosowano r żne testy w odniesieniu do nocycepcji termicznej, nocycepcji chemicznej i zachowania spontanicznego. W obecnej pracy oceniono, że kompleks posiada jon Co(II) skoordynowany poprzez 4 i 5 pozycję flawonoidu. Metal wiąże się z flawonoidem w stosunku 1:2, co zostalo potwierdzone metodą Jobsa. Nasze wstępne dane dotyczące latencji odpowiedzi w testach hot-plate i tail- flick sugerują, że systemowo podawany CNC wywoluje znaczący efekt przeciwb lowy w warunkach termicznej stymulacji nocyceptywnej. Test spontanicznego zachowania wykazuje znaczący spadek aktywności.

Es wird angenommen, dass die Aktivit ten von Flavonoiden zunehmen, wenn sie mit bergangsmetall-Ionen koordiniert werden. Es wurde ein Versuch unternommen, den Beitrag der Metallkoordination und die Art der Wechselwirkung zwischen einem Flavonoid und einem Metallion zu verstehen und seine biologische Aktivit t zu bewerten. In dieser Studie wurde ein neuer Metallkomplex von Co(II) mit Naringin synthetisiert & durch FT-IR, UV-VIS Spektrophotometer charakterisiert. Gruppen von sechs M usen wurden mit dem Kobalt-Naringin-Komplex (10 mg/kg K rpergewicht) behandelt, der IP verabreicht wurde. Verschiedene Tests wurden verwendet, um die antinozizeptive Wirkung oder den Einfluss auf das Verhalten der getesteten Substanzen auf thermische Nozizeption, chemische Nozizeption & spontanes Verhalten zu bewerten. In der aktuellen Studie wurde ausgewertet, da der Komplex ein Co(II)-Ion hat, das ber die 4 und 5 Position des Flavonoids koordiniert ist. Das Metall bindet mit dem Flavonoid im Verh ltnis 1:2, was durch die Jobs-Methode best tigt wurde. Unsere vorl ufigen Daten f r die Reaktionslatenzen bei Hot-Plate- und Tail-Flick-Tests deuten darauf hin, dass systemisch verabreichtes CNC eine signifikante analgetische Wirkung unter thermischer nozizeptiver Stimulation erzeugt. The activities of flavonoid are believed to increase when they are coordinated with transition metal ions. An attempt was made to understand the contribution of the metal coordination & type of interaction between a flavonoid & metal ion & evaluation of its biological activity. In this study a new metal complex of Co(II) with naringin was synthesized & characterized by FT-IR, UV-VIS spectrophotometer. Groups of six mice were treated with Cobalt Naringin Complex (10 mg/kg body weight), administered IP. Different tests were used for evaluating the anti-nociceptive effect or the influence on behavior of the tested substances on thermal nociception, chemical nociception & spontaneous behavior. In the current study it has been evaluated that the complex has a Co(II) ion coordinated via 4 and 5 position of the flavonoid. The metal binds with the flavonoid in the ratio 1:2, which was confirmed by Jobs method. Our preliminary data for response latencies for hot-plate and tail- flick tests suggest that systemically administered CNC produces a significant analgesic effect under thermal thermal nociceptive stimulation. The spontaneous behavior assay shows a significant decrease of activity.

In molti paesi ci sono poche linee guida di medicina obsoleta, ma l'OMS coordina una rete per ispirare un uso sicuro ed equilibrato. La ricerca farmaceutica rimasta attenta nel trovare usi obsoleti delle piante medicinali nel suo duro lavoro di rilevamento dei farmaci.Gli studi tecnici per capire l'effettiva efficacia e gli altri limiti di questi farmaci ne amplierebbero assolutamente le possibilit di utilizzo futuro, se si rivelassero davvero efficaci.
